Biochemical and morphological changes during the growth kinetics of Araucaria angustifolia suspension cultures BABT
Santos,André Luis Wendt dos; Silveira,Vanildo; Steiner,Neusa; Maraschin,Marcelo; Guerra,Miguel Pedro.
Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. For both the embryogenic cultures a preferential uptake of glucose in the late stages of cell growth kinetics was observed. The extracellular protein content was similar for both the embryogenic cultures. Acetocarmine and Evan's blue double stain showed major differences for early somatic embryo organisation, in which only the embryogenic...

Botanical source investigation and evaluation of the effect of seasonality on Brazilian propolis from Apis mellifera L. Scientia Agricola
Tomazzoli,Maíra Maciel; Zeggio,Amélia Regina Somensi; Pai Neto,Remi Dal; Specht,Leandro; Costa,Christopher; Rocha,Miguel; Yunes,Rosendo Augusto; Maraschin,Marcelo.
ABSTRACT: Bees generally use different botanical sources of resins for the production of propolis. The elucidation of botanical sources of propolis and identification of the effects of seasonality on the chemical profile of propolis are recognized as two important aspects in the development of a high quality product. Thus, our objective was to elucidate the botanical source and identify the effect of the seasons on the chemical profile of propolis produced in southern Brazil. The chemical profile of the samples was analysed by spectrophotometric and chromatographic techniques and the results were coupled to multivariate analysis. Field observation of the foraging behaviour of Apis mellifera L. revealed its preference for the Baccharis dracunculifolia DC....

Effects of salinity on the physiology of the red macroalga, Acanthophora spicifera (Rhodophyta, Ceramiales) Acta Botanica
Pereira,Débora Tomazi; Simioni,Carmen; Filipin,Elisa Poltronieri; Bouvie,Fernanda; Ramlov,Fernanda; Maraschin,Marcelo; Bouzon,Zenilda Laurita; Schmidt,Éder Carlos.
ABSTRACT Salinity is an important abiotic factor since it is responsible for the local and/or regional distribution of algae. In coastal regions, salinity changes with prevailing winds, precipitation and tide, and particularly in extreme intertidal conditions. Acanthophora spicifera is a red seaweed that occurs in the supratidal region in which changes in abiotic conditions occur frequently. This study evaluated the effects of salinity on the metabolism and morphology of A. spicifera. Algae were acclimatized under culture conditions with sterilized seawater for seven days. Experiments used different salinities (15 to 50 psu) for seven days, followed by metabolic analyses. This study demonstrates that extreme salinities affect physiological parameters of A....

Genetic variability for carotenoid content of grains in a composite maize population Scientia Agricola
Kist,Volmir; Albino,Vanessa Stainbach; Maraschin,Marcelo; Ogliari,Juliana Bernardi.
Local maize (Zea mays L.) varieties are cultivated by small-scale farmers in western Santa Catarina (SC) State, in southern Brazil. These small areas frequently present many problems related to biotic and non-biotic stresses, which have limited the economic output and income of the farmers. Production from local varieties for human consumption would be an alternative way of improving income and stimulating on farm conservation. The genetic variability of the total carotenoid content (TCC) of kernels in a local maize population was evaluated for their economic exploitation potential as biofortified food. Two independent samples of 96 half-sib families (HSF) plus four checks were evaluated in two groups of experiments in western SC and each one was carried...

Hypnea musciformis (Wulfen) J. V. Lamour. (Gigartinales, Rhodophyta) responses to gasoline short-term exposure: biochemical and cellular alterations Acta Botanica
Ramlov,Fernanda; Carvalho,Tiago Jose Griebeler; Costa,Giulia Burle; Rodrigues,Eva Regina de Oliveira; Bauer,Claudia Marlene; Schmidt,Eder Carlos; Kreusch,Marianne Gabi; Moresco,Rodolfo; Navarro,Bruno Bachiega; Cabral,Debora Queiroz; Bouzon,Zenilda Laurita; Horta,Paulo Antunes; Maraschin,Marcelo.
ABSTRACT Presence of toxic compounds in marine coastal waters has increased exponentially since Industrial Revolution. In this way, we aimed to evaluate biochemical and physiological changes occurring within Hypnea musciformis after short-term exposure to gasoline. Hypnea musciformis was cultivated without gasoline and then exposed to various concentrations of it (0.001 % - 1.0 %, v/v) for periods of 30 min, 1 h, 12 h and 24 h. A Pricncipal Compound Analysis of UV-vis spectral window (200-700 nm) was able to discriminate gasoline-exposed samples according to both exposure time and gasoline concentration. Changes in carotenoid profile composition were observed. Decreased carotenoid content was associated to gasoline exposure time, being lutein and...
